James Hamilton, 1st of Raploch, who got a charter of the lands of Raploch from James, Lord Hamilton, in 1440, wherein he is styled " dilectus consanguineus suus"
All male line descendants of this person will be the SNP A376, that is just the way genetics works. The FTDNA Hamilton Surname project group recognizes the Raploch Hamiltons as Group A Hamiltons with a terminal SNP of A376. More advanced SNP work has further split this group into several sub-groups, see the tree maintained by YFULL for a detailed breakout of the haplotree under A376.
For an explanation of Y chromosome testing for genealogy purposes see Wikipedia at this link.
Another rendition of the haplotree can be found at the ISOGG tree and the current haplogroup designations against specific SNPs can be found at this link.
Note STR patterns can be used to predict a haplogroup whereas specific SNP testing accurately defines a haplogroup.
