JACKSONS OF KENTUCKY: THEIR ANCESTORS AND DESCENDANTS lists sons Ralph, Francis, Joel and Lewis.
THE JACKSONS OF LOWER VIRGINIA suggests sons Joseph, Joel, Francis, Lewis, possibly Mark.
These articles have identified a Patience married to Ezekiel Sudbury as the possible daughter of Ralph's brother Joseph. This is an erroneous connection - Patience, daughter of Joseph, would not have been old enough to fit the records found, and she is believed to have died unmarried. It is possible this Patience is instead a daughter of either Ralph or Daniel, most likely Ralph since his is older. Both Ralph and Daniel are thought to have had daughters, names unknown. Here are the records of Patience Sudbury, wife of Ezekiel Sudbury:
16 Aug 1746. Mark Jackson, son of Williams, sold 130 acres to Thomas Franklin Jr, part of 490 acres patent to Williams Jackson on 15 Mar 1735 in Henrico County on Licking Creek. Deed was witnessed by Ralph Jackson, John Stewart, John Green, Patience Sudbury, & Martha Jackson
1746 Henrico Co Deed. Ezekiel Sudbury gave a deed of gift to Ezekiel, "son of my wife Patience" Witnesses were Martha Jackson, John Green & Ralph Jackson. [No exact date given, but needs to be examined since Patience, wife of Ezekiel, seemed to be living in August to sign the previous deed. If indeed Patience the wife was already deceased in August, then the deed may have been signed by Patience, daughter of Ezekiel, making the wife even older and perhaps placing her in a previous generation.]
8 Apr 1747. Henrico Co Deed. Ezekiel and wife Anne gave a deed of gift to Ezekiel, his son by his wife Patience, dec'd, and to his daughters Hannah & Patience. Witnesses were Francis Jackson, Thomas Sadler & Stephen Beasley.
Note: There is nothing here to suggest Patience, wife of Ezekiel Sudbury, was a Jackson except the existence of witnesses by that name. She may be someone else entirely.

DNA Connections
It may be possible to confirm family relationships with Ralph by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA. However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line. It is likely that these autosomal DNA test-takers will share some percentage of DNA with Ralph:

Amanda, On Ralph I's profile, it shows two Ralph's as sons. There was only Ralph m. Obedience___ . The second Ralph is stated as born 1721 which would be difficult for someone who died in 1708/9. I'm going to unlink him. It says he married a Mary Clegg? The only Ralph I know of after Ralph II, is Ralph III, son of Ralph II( aka Ralph of the Cottage) b. abt 1714 died bef. 1776 but in deeds, his wife is Hanna. There is one deed with a Martha, but I'm not sure if that deed is his of the Ralph, son of one of Ambrose's group. At times it gets hard to keep them apart. I have only listed the children of Ralph I and Ralph II that I can prove through deeds, since the first died without naming male children except "My sons" and the second did not leave a will, having deeded land to each child. Both Ralph I and Ralph II had daughters. Ralph I had Elizabeth (Perkinson) and Ralph II had Pheby (Powers). Ralph III died intestate and an inventory was taken in 1776. He seems to have the following but I would not swear to it=by Hanna: Phebe, Simon, By Martha: Catherine, Lucy, Edith, Martha and Elizabeth.
