DNA Connections
It may be possible to confirm family relationships with Ann by comparing test results with other carriers of her ancestors' mitochondrial DNA. However, there are no known mtDNA test-takers in her direct maternal line. It is likely that these autosomal DNA test-takers will share some percentage of DNA with Ann:

The mistake is not the poster's fault, but mine. I omiitted the word Patterson as part of Josiah's name in transcription of Ann's will. The long blank space before the name Davidson is for the name Mary Neil, daughter of James Holmes and Ann and married to John Davidson. Since James Holmes's first wife was Mary Neill, people usually assign motherhood of Mary Neil Holmes to her, but she was already dead at that time. Ann and James may have named the daughter for the first wife, who was Ann's friend, or for Ann's sister Mary (Alexander) Neill, or both.

The father of Ann's father John Alexander, married to Rachel Davidson, was James Alexander as shown by YDNA evidence and deeds made by John to his sons and one daughter shortly before his death in 1753. James's wife was named Ann (maiden name unknown to me).
