Anthony Ashley-Cooper was born on 17 September 1761, the eldest son of Anthony Ashley-Cooper, 4th Earl of Shaftesbury and his second wife, Mary Bouverie.
He succeeded his father as Earl of Shaftesbury on 27 May 1771. His wife was Barbara Webb whom he married on 17 July 1786.
Anthony died on 14 May 1811 aged 49. Having no son he was succeeded as Earl of Shaftesbury by his brother, Cropley.
