Based on Big Y testing at FTDNA.com Benjamin Carter (abt.1700-aft.1754) was William Carter's uncle.

Possible parents are William Carter b. 1628 and Elizabeth (unknown) Pettway Carter b. 1639.
William who married Elizabeth (as mentioned in Robin's sidebar comment) and Joseph who married Mary Grant could be some sort of relative of this William. All 3 had marriages in the 1680s. Joseph could be a son or brother, but William would have to be a son or some other relation. If Joseph and/or William were sons by an earlier wife there is no indication of such in the known records. Still, it seems reasonable to consider their presence there could provide clues.
Maj. Thomas is the only Carter known who was a landowner there earlier but he didn't name any children as his heir. While not proof of anything, it should be noted that William's first son by his first known wife was named Thomas.
Locating the land that William owned and lived on might help too. Abr. Weekes was upriver of Urbana and William Churchill was down river from the church. Haven't located Geo. Goodloe yet either.
Most of the headright land grants seem to be for William of Surry, but this one looks interesting. It's only about 15 miles or less from Middlesex Co.:

Bet. some of the head branches of Pepetick Cr. & Mr. Popes Cr., beg. near Rappa. path, extending to the head of lands of Silvester Thatcher & Thomas Whitlocke & adj. land of Thomas Wright. Trans, of 18 pers: ... Wm. Carter, ....[15]
There was an earlier, probably older, William in Lancaster Co. before Middlesex Co. was formed. He apparently died there by 1664 and left a daughter Susan who was under 20. (If he had any sons they were likely adults as none are mentioned.) Susan was assigned first to John Edwards, then Thomas Marshall (who was both a neighbor of and handled affairs for Col. John Carter) and reassigned to Richard Merryman.[16] p. 34, 63 (can't post abstract due to strict copyright). This older William could be the one mentioned in the headright claims above. Relationship to other Carters of Lancaster Co. is unknown, but the possibility of him being the brother of John (and Maj. Thomas if he is also a brother) should be considered.
Broken Branches are the most distant known ancestor (aka "brick walls") of men who have a Y DNA test that they are descendants of William Carter.
20 Feb 2024 Goodloe Carter, b. 20 Sep 1784, Albemarle, VA d. 7 May 1848, Woodford, KY Parents were determined to be George Carter (abt.1723-) and Frances Diana Goodloe (1764-) thanks to research by Paul T Carter.
