It may be possible to confirm family relationships with John by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.

I believe he is actually the brother of Jacob and not the son of John. There is no record of a son John born to Jacob's brother. More likely the John baptised 28 Jul 1611 in Heptonstall, Halifax, Yorkshire, England is the man who married Joanna in Woburn and was the father of her 7 children.
