DNA Connections
It may be possible to confirm family relationships with William by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA. However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line. It is likely that these autosomal DNA test-takers will share some percentage of DNA with William:

There are some inconsistencies in the profiles for Thomas Holder and Susannah Bunch, but most family histories put them in eastern North Carolina (Bertie County) or adjacent Virginia. Culpeper VA is considerably further north. What is the evidence that William was born there?
There is a will for William Holder in Orange County, NC, probated 5 May 1817. See North Carolina, US, Wills and Probate Records, 1665-1988 (Orange>Wills, Vol E-F, 1822-1851. The inferred Death Year is listed as abt 1817. The will mentions his wife Mourning (which matches a marriage record for Granville Co with Mourning Duggar) and the following children: Chesley, William, Daniel Polly, John, Hosea, Lydia, Nancy Elijah, Elisha and names Chesley as executor. This appears to me to be enough information to update the death date and children, but note that Gabriel is not listed in the will. I'm not sure of the source that exists for Gabriel being a son. Take a look and see what you think. Thanks. Addition: I have since seen a transcription of a second will of William Holder, dated 15 May 1817, and transcribed by Ben Franklin of Durham, NC, and sourced as: Original Will, on file in Raleigh, NC Archives CR 073.801. The two wills are very similar and this one also refers to Mourning as his wife and the children: Chesley, William, Daniel, Polly, John, Hosea, Lydia Nancy, Elijah and Elisha.
