Sir Alexander Mackenzie was a Scottish explorer. He is known for his overland crossing of what is now Canada to reach the Pacific Ocean in 1793. This was the first east to west crossing of North America north of Mexico and predated the Lewis and Clark expedition by 10 years.

DNA Connections
It may be possible to confirm family relationships with Alexander by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.
However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line.
It is likely that these autosomal DNA test-takers will share some percentage of DNA with Alexander:
Alexander may have had three siblings: Murdoch, Margaret and Sybilla.
Margaret is conjectured to have married James Dowie (sic), On her grave memorial in Greyfriars Private Cemetery he records: "This Stone is Erected by Capt James Douie of the Wellington Revenue Cutter, in gratefull and affectionate Remembrance of Margaret MacKenzie his Spouse who died April 1815 &c whose Remains are here inter'd".
Geddes Margaret Mackenzie (Alexander's daughter with his Scottish wife, Geddes Mackenzie) was also memorialized on the same stone https://www.gravestonephotos.com/public/gravedetails.php?grave=138555
In his will James states his wishes thus: "assign convey dispose and make over to and in favor of Mrs Mackenzie or Dowie my Wife, Sir Alexander Mackenzie of Avoch Knight my Brother in Law..." https://discovery.nationalarchives.gov.uk/details/r/D267232
Contact with Stornoway Historical Society may provide information about Alexander's siblings.
Information about his two children to Metis, Julie and Andrew might be had from article and correspondence between Judy Parry and Pia Goddard in the September 2006 edition of the Clan Mackenzie Canada Newsletter, http://www.electricscotland.com/mackenzie/images/news40.pdf

There are extensive relationships and marriages between Downie and Mackenzie families in Stornoway in the late 18th C. It is not unusual that Margaret Mackenzie might have married James Dowie. A marriage record has not been identified
This requires further research.
