It may be possible to confirm family relationships with Amasy by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.
However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line.
It is likely that these autosomal DNA test-takers will share some percentage of DNA with Amasy:
Given that William Maston Mullins spent time living first in Denton County, TX, and later in Erath County, TX, I have spent some time trying to locate potential relatives among the residents of these counties. I have found one Sarah Elizabeth (Mullins) Haggard (Mullins-3877), who lived Denton County and seems to be descended from residents of eastern Kentucky. I have also located Catherine (Mullins) Pilkenton (Mullins-3754), seemingly buried in Erath County, TX. Her father appears to be from Lawrence County, Missouri (WM Mullins was born in Missouri), and there seems to be circumstantial evidence also linking her family to eastern Kentucky, possibly leading to William M Mullins II (Mullins-2469).
It seems likely that William Maston Mullins (buried at Rock Creek Cemetery), and William Mullins son of Flower and Lucretia Mullins, are, in fact, different people. I come to this conclusion due to a disagreement between the dates of birth, and the following census record on family search: https://familysearch.org/ark:/61903/1:1:M6ND-NR8
