Biography
William was the oldest son of Henry Shelton (abt.1738-bef.1799). He married Fanny Maupin, daughter of John Maupin on 15 Aug 1783.[1] William was named as executor of his father's will in Aug 1799[2] which was proved in Dec 1799 in Albemarle, Virginia.[3][4]
On 3 Sep 1804, brothers William, Austin, and Thomas Shelton sold their inherited property from Henry Shelton to James Barbour. Excluded was land intended for the Smith children (Susannah's).[5]
On 8 May 1809, William sold land that he had inherited from Henry Shelton to Bazaleel Brown. His wife, Fanny, was required to relinquish her dower right.[6]
Research Notes
Some have concluded that this William is the son of William Shelton (abt.1730-bef.1803). This may be based on William Shelton being a bondsman for the marriage to Fanny Maupin. However, it is the son of Henry Shelton (abt.1738-bef.1799) who married Fanny. This is proven in an 1809 land transaction, where William Shelton, identified as executor of Henry Shelton, sold land. In Henry's will, William was identified as his son and executor. In the land sale, William's wife Fanny was required to relinquish her dower rights. This would have only been necessary if William owned the property inherited from Henry.
Some have also concluded that William had a first wife named Lucy Harris. This is also incorrect as Lucy was the wife of the aforementioned William Shelton (abt.1730-bef.1803) and named in his 1803 will.

It may be possible to confirm family relationships with William by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.
However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line.
It is likely that these autosomal DNA test-takers will share some percentage of DNA with William:
Shelton-2126 and Shelton-928 appear to represent the same person because: As I posted in the comments of Shelton-2126, the William Shelton who married Fanny was the son of Henry, not William. I just adopted Shelton-928 and will work on additional sourcing.
The William Shelton who was the husband of Fanny Maupin was the son of Henry Shelton (abt.1738-bef.1799). Henry Shelton's will named his "beloved sons William & Austin Shelton as sole executors." In 1809, William, as executor of Henry, sold the land (https://www.familysearch.org/ark:/61903/3:1:3Q9M-CS4H-8WQG-P?i=298&cat=283051). Most importantly, his wife, Fanny, had to relinquish her dower right. That confirms that the land was inherited by William from Henry as that is how Fanny would have acquired Dower rights. That also means that William was still alive in 1809. I will be proposing a merge based on the above.
