However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line. It is likely that these autosomal DNA test-takers will share some percentage of DNA with William:

There is pretty good sourcing for this William Sims-105, as well as his parents, Wm Sims-36 & Martha Ryan-4928, as well asa significant number of the direct descendants.

I have just gotten a start on them with daughter, Mary Sims-3123. go to the online source family history for more names and please add profiles.

I can't source or provide evidence of Elizabeth Howard being a spouse of Sims-36. there was also some possible confusion with Howard being mother of children of Wm Sims-99.

If Howard was 1st wife of Sims-36 that should be identified with the specific children-descendants. there should be records Culpeper, of those events.

Sims-36 marriage to Martha Ryan-4928 is given as early 1754 let me hear if you find something

Annals of the Family of John Sims, by his son C. N. Sims, was published in 1893, and is available in a digitized version on the web. William Sims, b. 1740, was the great-great-grandfather of the author.
