It may be possible to confirm family relationships with Robert by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.

The sons of Robert Tucker-299, Joseph Tucker and James Tucker-2010, have a YDNA match with zero mutations. A YDNA comparison should be done for Robert Tucker-299 and Thomas Tucker-300. The Find A Grave Memorial and Norma Tucker's book Colonial Virginians and Their Maryland Relatives and Ancestry are questionable. It is difficult separating the many Robert Tuckers living in the same area and time period. Family Tree DNA has many DNA tests for Tucker and someone with YDNA knowledge should look into the connection for this Robert and father Thomas. The Family Tree DNA tests are listed by area. The Charles City County group has the largest amount of testers (90 tests) and there are no matches for a father of Robert Tucker-299. Thomas Tucker-300 is most likely part of one of the Maryland testing group.
The only good way to separate lines using dna is a Phylogenetic Tree using FTDNA Big Y results. In this manner, you can separate the different Tucker lines very effectively.
Having a Ydna match even at 111 markers is still not definitive enough for genealogy.

The Big Y test at FTDNA is not the same as a Phylogenetic Tree. Check it out. We did this in the Big Y Gresham Surname Project and it made a significant difference.
I am assuming that you have looked at the Tucker lines here on WIKI and made notations when they do not seem to be charted according to the Haplog groups in the Big Y Block chart. Good work! Mary
