Is it desirable that these should always be set, one way or another?  Or is there a place for leaving them unset?

I think this is what they are discussing here, it might relate to your question

According to Help:Relationship Status:

"Selected indicators will appear on certain family tree views and will affect Relationship Finder results."

So while I don't believe it is a hard requirement to set them, I would say it is a best practice to do so.

But the settings make no difference to the Relationship Finder.
But they do mark the relationship as confident or uncertain, etc.

They should be set when you are confident. These status buttons are one of the features that make WikiTree data that more reliable to others.  In comparison to Ancestry.com, I would have to use a TreeTag to accomplish the same effect.

Everyone should make an asserted effort to set the statuses on each profile as appropriate.
