 I finally made contact with a niece of my said father's and her first response was DNA test!

But I assume you ask what test to take.... Family Finder the autosomal test is the normal test to take see http://www.wikitree.com/g2g/242851/which-dna-test-would-be-best 

If you select DNA in the drop down menu you will find

1. DNA Ancestor Confirmation Aid for Jo-Anne (Reid) Zeron
   1. I assume person nr 8 is the common ancestor 
      Willard Harrison Reid (Reid-5431)  
      1. relation great grandfather
      2. descendants
2. DNA Ancestors

My understanding is that this weekend it's cheaper to test at FTDNA 79$

Since you are both female, you want the autosomal DNA test, either AncestryDNA from Ancestry.com or Family Finder from FTDNA. You both need to use the same company!

I prefer FTDNA, but there is an advantage to using Ancestry, if you use Ancestry you can transfer your results later to FTDNA at reduced cost, which allows you to research in both databases. Only FTDNA accepts transfers, so if you think you might be interested in this test first at Ancestry.

Whichever way you go, once you both have your results you'll want to upload your raw data to the GEDMatch website; it is a free site that allows you to do a number of analysis routines and compare your results to many others'.

If she IS the niece of your father you should known almost immediately, you'll share a sizable chunk of DNA.  Good Luck!

It would be easier to use the same company, but if perchance that doesn't work out, there's gedmatch. Heck, even if you do use the same company, you would benefit from uploading your results to getmatch if you're interested in finding cousins.

Yes, you could use different companies, I over-emphasized my opinion that using the same is preferable. It is a tad less complicated to compare your results if they come from the same source, but you can still compare if you use either, and uploading to GEDMatch is recommended regardless.

To comment further on this - If you test with Ancestry then you will have your DNA in that large Gene pool. Then as stated, you can transfer your raw data file over to family Tree DNA for about for $40.00US. This way your DNA will be in TWO big Gene pools. Adding it to GEDmatch will add it to yet another large Gene pool. DNA.Land would be another Gene pool too. The last two are free.
