Can autosomal DNA test confirm 6th generation grandmother via males?

Broken connection to my Ouellette line due to destroyed church records. Trying to confirm broken link by proving relative of Marie Genevieve Tardif married to Jean Baptiste Ouellet. I believe she is my 6th great grandmother and want to confirm this via the male line. We know we are related to the Ouellet's from La Pocatière in Quebec but not sure which line and that is why we want to focus on Jean Baptiste's wife.

For autosomal DNA whether it is a male or female line doesn't matter. For a 6th gt grandparent the percentage is so small (about 1.6%) that it could very well not matter.
For confirmed by DNA you will need at least 2 confirmed (not by DNA) descendants of Marie to have tested and you to have shared a specific piece with them. (Not necessarily the same piece, however).
