No, no more that if his name had ended in Jr.

The II part (or jr) refers to his fore names, not his surname. All it means is that his father was called John Henry Hempel. His wife would only take the Hempel part.

I agree with David, but I have to ask another question:

Why does Wikitree put the extension on the wife's surname?

Hello, Jon!

The reason Vanilli's profile shows the II in her (married) surname is that it is listed in her Current Last Name field. Just go in and edit that field to correct her name display! The II is not part of her - or her husband's - surname.

Lindy is correct. - Looking at the profiles...  John Henry Hempel II correctly has the 'II' as a suffix. When you entered Vanilli's Current Last name, you added the 'II' as part of it. Just edit it out and all will be correct.
