I have a Ancestry DNA test from my father's brother, would that be useable?

Yes, it's certainly usable. Attach the details of the test to your uncle's profile, make sure his profile has at least a semi-private profile and it will be propagated in the tree. His parents will need to be semi-private also.

P.S. semi-private, to me, is a yellow or apricot privacy setting.
