Joan was born about 1510, the youngest daughter of Thomas Berkeley and his wife, Eleanor. On 24 June 1527 she married, at Yate, Nicholas Poyntz, son of Sir Anthony Poyntz of Iron Acton in Gloucestershire. She bore him nine children, Sir Nicholas, Francis, Edmond, Anthony, William, John, Anne, Jane and Frances. Her husband made his will on 26 February 1556 and died on 27 or 28 November.
Joan took as her second husband Sir Thomas Dyer and bore him three sons. It is said that his ill-treatment of her caused her death.
Smyth, usually reliable when working from legal documents, lists the issue of Joan and Nicholas Poyntz in some detail. However, regarding her second husband, he merely remarks that ' .....in her Elder years marryed to S' Edward Dyer, and dyed in the fixth of Queen Elizabeth.' but without providing a source.
