Research Notes
There seems to be some doubt on whether the Mary that married Richard Estes was indeed Mary Yancy. She is said to have been a daughter of Charles Yancy, but the relationship is not proved.
On 23 April 1747, after the death of her husband, the vestry of Fredericksville Parish employed Mary Estes as sexton of the church at North East Creek and allowed her wages of 500 lbs. of tobacco. On 28 July 1747, she indentured her son Charles, a poor boy, to learn the carpenter trade.

It may be possible to confirm family relationships with Mary by comparing test results with other carriers of her mitochondrial DNA.
However, there are no known mtDNA test-takers in her direct maternal line.
It is likely that these autosomal DNA test-takers will share some percentage of DNA with Mary:
I found this interesting: Check out - "A Tapestry for Elizabeth: The Story of the Ancestors of Mary Elizabeth Hoffman", By Thomas Paul Merrill, Page 93.
"Samuel Alexander Saxon was born 1699 in Rockdale, Lancaster, Hampshire, England and died in Halifax County, N.C. in 1766. He married Mary Yancey in 1722. She was the daughter of Charles Yancey. She was born 1703 in Rockdale, Lancaster, England. She died in 1770 in Halifax County, N.C."
Next page goes on to describe their living adjacent to "Acheleus Yancey, son of Charles Yancey" (dunno why they don't just say 'Elizabeth's Brother'.

There is definitely another Mary Yancey that marries Saxon, but... I can't tell from whom this one comes:
