DNA Connections
It may be possible to confirm family relationships with John by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA. However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line. It is likely that these autosomal DNA test-takers will share some percentage of DNA with John:

John (Jr)'s children appear to be: Caroline, Betsey S., Mary J., William S., Angeline R. "Ann". On the 1850 census in Farmington, Franklin, ME, John, wife Mary, and Ann appear, along with an Edmund. He is not mentioned as a child of John Jr. in the book on the Dow family. The year of birth of this Edmund closely resembles that of John Jr.'s brother Edmund's son, Edmund (Jr). It's also not known if Mary L. Smith had children from her previous marriage that John may have adopted. Edmund's presence on the household census, without notation of relationship, merits his being considered speculatively as a son.

John Jr. is believed to have married a widow named Mary Love Smith. He may have earlier been married to an Elizabeth Saley/Salley. See: daughter Betsey's death documentation; son William being referred to as having the middle name "Salley" (documentation?). Of curious note, An Elizabeth Saley married an Ebenezer Thompson, both being buried in Pease Cemetery in Avon. One of their children, Susannah Thompson, was mother to Isaac Clark, husband of John Jr's sister Mary. It appears that "Saley" then was a surname present in that area at the time, and they had a familial relationship to John Jr.'s sister Mary. All of this taken into consideration, it's possible John Jr. may have been married twice.

John and Mary are present in Weld in 1860. It appears John Jr. died on 30 Nov 1864 and was buried in Thompson Cemetery.

Side note: I descend from John Jr's daughter Elizabeth/Betsey S. Dow Billington. Ancestry.com DNA suggests that Angeline R. "Ann" Dow is a half sister to Betsey, sharing a common father, which lends further to the possibility John Jr. married twice. However, it appears the DNA tested descendant of Angeline is also related to the Billingtons, specifically, through Betsey S. Dow's husband's sister, Locada Billington.
