DNA Connections
It may be possible to confirm family relationships with John by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA. However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line. It is likely that these autosomal DNA test-takers will share some percentage of DNA with John:

To my knowledge there is no evidence at all that links John Elswick to Henry Thomas Elswick and Susanna Hampton Elswick as parents.

See the letter from the Rev Albert Elswick quoted in the sources above about John Elswick and wife Rachel: “They appear out of nowhere, receiving a 180 acre land grant near Sharpsburg, MD. in 1742.”

At the moment John Elswick and Rachel appear to be a brick wall.
