DNA Connections
It may be possible to confirm family relationships with John by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.
However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line.
It is likely that these autosomal DNA test-takers will share some percentage of DNA with John:
After a bit of poking around, it looks like this is John's first wife: Ruth (Chamberlain) Fletcher, probably mother of John the younger. Nathaniel Chamberlain, mentioned below, was her father, and his estate was administered by wife Abigail in 1716. The estate was finally divided in 1723 after Abigail's death, under Benjamin Chamberlain (son, also noted on the guardianship papers previously mentioned) by which time Ruth was deceased, John was remarried. Voila. See Plymouth County Probate case #3727.
Misc. finding... was checking probate records and found in Plymouth Probate Records, case #7800, a guardianship record where John in Oct 1723 in Harwich, as guardian to son John, recieved from the estate of his grandfather Nathaniel Cha[se? /Chamberling? page torn] of Pembroke £11.
