Third son of Lieutenant General James Hamilton and his wife the Hon. Elizabeth Gerard.
Anne was named for his Godmother, Queen Anne.
Hamilton married Anna Charlotta Maria Powell in October 1742. They had two sons.
Hamilton also had an illegitimate son with Mary Edwards, daughter of Francis Edwardes.
Hamilton was commissioned into the Coldstream Guards between 1731-1733.
He died in France in 1748 and is buried at St James's Church, Piccadilly.
