Help:Advanced DNA Confirmations

DNA Confirmation using an X-chromosome or mitochondrial DNA match is more complicated than Y-chromosome confirmation or a one-to-one autosomal confirmation for a close cousin. There is also less agreement in the WikiTree community about the requirements. To discuss any of this, post in G2G using the tag DNA.
As an alternative to following these instructions, you can use Greg Clarke's DNA Confirmation app.
X-Chromosome DNA Confirmation
If the matches are both men there is a special opportunity for one-to-one confirmation if they match on their X chromosomes. Since men have only one X chromosome, no triangulation is necessary.
Here is an example of what to put in the Sources section of the profile when you mark a person's father or mother as Confirmed with DNA using xDNA:
- * Maternal relationship is confirmed by a 108.0 cM X chromosome match between John Kingman and his second cousin once removed Kelly Miller on GEDmatch. Their most-recent common ancestor is Martha (Lanning) Babst.
If a match is not on WikiTree and is living do not include their name. Use their initials or another anonymous identifier.
Mitochondrial DNA Confirmation
If two people are an exact mtDNA match on HVR1 and HVR2 (not counting minor mismatches) then their direct maternal lines back to their most recent direct maternal line ancestor is confirmed.
Here is an example of what to put in the Sources section of the profile when you mark a person's mother as Confirmed with DNA using mtDNA:
- * Maternal relationship is confirmed by an exact HVR1 and HVR2 match between this Family Tree DNA mtDNA test for Priscilla Weatherford and this Family Tree DNA mtDNA test of her maternal line cousin Clair Zimmerman.
If a match is not on WikiTree and is living do not include their name. Use their initials or another anonymous identifier.
