Research Notes
In 1840 US Census, he may have been living in Tompkins Co., Hector, New York. There is a William Jefferson listed with a male child between 5 and 10 years of age (my maternal great-grandfather, Daniel Jefferson, the son of William, would have been about 7 years old in 1840).
In the Iowa Census of 1886, this William Jefferson is listed as 57 years old, and his wife Johanna, is listed as being 57 years old and he is a hotel keeper. His son, Daniel is also listed as a hotel keeper. His daughter-in-law, Susanna, is also listed as being in the same household.

Note: I did a google search on William Jefferson today (January 22, 2015). It looks like we share this ancestor. My William Jefferson was in Mentz New York with his wife Joanna in the 1850 census that you referenced. My great grandmother Mary Adell Jefferson Crawford and my great grandfather Dewitt C. Crawford are also listed in that document. In the 1860 Iowa Census Olive (Jefferson) and DC (DeWitt Clinton)Crawford are listed with their daughter Mary Adell next door in the previous household. I have a little further information from a list of marriages births and deaths copied from a family Bible (I don't know who had it) probably by my grandmother. William's wife was listed as Joan Carpenter, their marriage as 11 Feb 1829 I found a newspaper item, which soon disappeared from the internet, which indicated that Daniel Jefferson died when struck by lightning 23 May 1860. I can send you a copy of the Bible entries if you are interested Mary Adele Taylor Wakefield, MA.
