Elisabeth Dee Prince was born in Holland, Michigan, on 8 January 1958. She is the daughter of Edgar Prince. She received her education at the Holland Christian High School and graduated from Calvin College with a bachelor of arts degree.

Betsy is currently married since 1979 as of 14 September 2020, with four children from her marriage. According to Forbes, her family was the 88th richest family in the United States with a net worth of approximately US $5.4 billion in 2016.

Politically, since 1982, Betsy is a member of the Republican Party. In 2004, she raised US $150,000 for the incumbent President's re-election campaign. During the 2016 election, Betsy endorsed three campaigns. As a political activist, she is known for her support of school choice, school voucher programs and charter schools.

On November 23, 2016, the then President-elect selected Betsy to be the Secretary of Education. The nomination received support and criticism. Betsy's nomination was approved by the Senate Committee on 31 January. On 7 February 2017, there was a 50-50 tie on the vote. The Vice President cast a tie-breaking vote to confirm Betsy's nomination as the United States Secretary of Education.

Betsy resigned as the Secretary of Education on 8 January 2021. One day earlier, she tendered her resignation to her president, saying on the storming of the Capitol on 6 January: "There is no mistaking the impact your rhetoric had on the situation."
