It may be possible to confirm family relationships with John by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.

I'm beginning to wonder if John William Ray hasn't got mixed up with another John William Ray. There is a John William Ray on FamilySearch who married Cynthia Lipencot, but whose parents are Samuel Ray and Nancy Caton. There is already a problem with this John William Ray's birth date; to line up with census data, it should be 1829, not 1833. Also, there was another child born to this family; John William has a younger sister, Elizabeth, who was born in 1833. These kind of relationship mix-ups aren't easy to detangle, but it can be done with patient research and some deductive reasoning.
According to the 1850 US Census for Sangamon County, IL, John Ray, was born in 1829. In 1850 he was 21 years old, living in the household of Samuel and Elizabeth Dillon. Also listed are Sela (Celia) Ray, age 19, Reason Ray, age 18, and Elizabeth Ray, age 17.
