It may be possible to confirm family relationships with Jacob by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.
However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line.
It is likely that these autosomal DNA test-takers will share some percentage of DNA with Jacob:
It has been determined that not only are this correct Jacob Souder and the other man, "Jacob W. Souder" of MD not the same person, Jacob "W." is not really valid...
Upon viewing of the actual MD deeds (not the transcription), of 1885/1887 that "Jacob W," signed, the "W" is actually not an initial...
It is his "Mark" for signature, and it is similar to a "W" but in fact is not a letter...it is an indication that he was illiterate, and did not use the usual "X" as his "mark." (Possibly because his wife Ann signed the 1887 deeds with an "X" for differentiation.)
In fact, the same and similar "mark" is used on the final will of a well researched Jacob Sowder (1734-1819) of VA leading to the possibility that he may be the same person who scratched his mark on the 1885/1887 MD deeds.
But the bottom line is, Jacob Souder (1865-1820) is not a "Jacob W, Souder" even if there was one...also, refer to the headstone photograph...no "W" in the inscription.
Jacob Souder (1765-1820) is NOT the same person as "Jacob W. Souder."
This has been a source of misinformation for a very long time. Jacob W. Souder is recorded on land deeds in MD as having a wife with a differing name at the same time as the confirmed wife of Jacob Souder (1765-1820).
In two of the sale transactions in 1887, this same man has a wife named Ann his seal is affixed to the name of “Jacob W. Souder.” One of the deeds states: “Aug 7 1787, Frederick County Court House, Frederick Maryland. Land Record Book WR-7 pages 405-406. Deed for 11 acres of land recorded in Frederick County, Maryland. Land that Jacob and Ann Souder sold to Ludwick Birely. At the request of Ludwick Birely the following deed was recorded August 7, 1787. to wit: This Indenture made this fifth day of March one thousand seven hundred and eighty seven by and Between Jacob Souder of Frederick County Farmer of the one part and Ludwick Birely of the other part.” This also identifies the “Jacob W. Souder” as being a farmer from Maryland…again, not a correct description of Benjamin’s brother Jacob (1785-1820) who was a resident of PA at that time.
Jacob Souder (1765-1820) married Elizabeth Rohrer (1765-1840) on May 1, 1787. He could not have been the same Jacob W. Souder who is recorded with the land deed also signed by Ann his wife on Aug. 7, 1787.

The son of John Souder (1726-1788) and Ann Bowman 1730-1794) should be Jacob Souder (1765-1820) which has been recently corrected and updated.
