- She was probably the daughter of his first wife, as Sarah Cosby, his widow, survived until 1740, which precludes the possibility of her having a child born to her prior to 1657.

- John married Sarh Higginson. We do not know about Sarah's parents; however, a Captain Robert Higginson had a daughter named Lucy Higginson, born 1620, who married Lewis Burwell. They are probably related.
- John Cosby owned 700 acres of land on the northeast side of Mattaponi River in New Kent County that he received for transporting sixteen [16] persons (date January 5, 1656). He received/bought 1,000 acres on the south side of the York River in New Kent County for transporting twenty [20] people, and another transaction is listed for 1,400 acres in New Kent County. So John Cosby was a big landowner in New Kent County.
- John Cosby and his wife Sarah had six [6] children: Charles, Mary, James, Margaret, Philip, and John. The son John Cosby, born about 1682, married Ann Stripling. The son Charles, born about 1670, married Jane Meriwether, the daughter of David Meriwether and Susannah Armistead and granddaughter of Nicholas Meriwether and Elizabeth Woodhouse.
- The father of John Cosby, born 1623, was Charles Cosby who was born in Stradbally Abbey, Queen County, Ireland, in 1575. Although Charles was born in Ireland, he was English. Charles's father, Alexander Cosby, a knight, and his grandfather, Francis Cosby, also a knight, had been chosen and sent by Queen Elizabeth to Ireland to rule Queens County, Ireland, and to own and manage the confiscated lands. The stories of their rule by the Catholic Irish are quite dramatic, bloody, and disturbing. Francis is said to have enjoyed hanging Catholic families from a large tree in front of his castle. Anyway, Francis and Alexander were killed in battle with the Catholic Irish. Alexander Cosby, John's father, was killed on Stradbally Bridge, in Ireland on September 19, 1596.
