DNA Connections
It may be possible to confirm family relationships with John by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA. However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line. It is likely that these autosomal DNA test-takers will share some percentage of DNA with John:

I'm really hoping to find out just how many children he (my fifth great-grandfather) had. An obituary for one of his sons (Samuel Calvin, born 1839) said he was the youngest of 14 children with Anna.

I also suspect he had children with two women in 1820 (Eliza with Catharine and America with Anna), and Ancestry ThruLines seem to confirm that.

i'm finding John Preston Spears and brother? George W. Spears b.1792 associated with the Peyton or Awbrey families of Virginia. Do any of those names ring a bell to you?

I know this John Preston does in fact have a brother named George, whose birth I have as Aug. 12, 1794 and in Northampton, PA. I don't have a death date or spouse for him. John's father's brother John Robert married Mildred "Molly" Peyton (1786-1850). Are you connected to the Spears bunch? We're also connected through the Babcock/Hillerich line.

I have opened up my Grandfather auto-DNA on Ancestry and detached the family tree from his results to see where the DNA hits would lead me. I already believed that g-g-g-grandmother Charity Peyton-1788 was a Peyton and according to DNA results to from her siblings and Aunts and Uncles it was a very strong correlation to his DNA. But there was also 2 connections of 13cm and 11cm from John and George Preston. Both the Peyton and Aubrey Familes are both from Northern Neck primarliy Loudoun and Fairfax Counties, which is pretty far from Cumberland County so i dont know if this is the shared DNA connection.

If the Spears family was as fruitful as they seem to have been, I wouldn't be surprised to see us related to most people in Virginia/West Virginia. I just looked at my ThruLines for John Preston and have 178 DNA connections. Those farmers could travel. When they made their way to Mason they linked up with the Cobb and Snodgrass families several times over as well, which has made color coding those DNA matches a lot of fun. I'm interested to see your 3 GGM is connected to my Price/Morton line as well, so you're connected to a lot of my Kentucky families!
