It may be possible to confirm family relationships with Robert by comparing test results with other carriers of his Y-chromosome or his mother's mitochondrial DNA.
However, there are no known yDNA or mtDNA test-takers in his direct paternal or maternal line.

Might be a good time to start digging through Hinshaws Encyclopedia for references to a Robert Stanley in VA, NC, & Indiana. There was a push in the early 19th century to establish new meeting houses in Indiana and many Quakers migrated from NC & VA. If he was in fact from the Cedar Creek Stanley line, clues may be found in the minutes of the Monthly Meetings.
It appears Nancy did actually die in 1856, as Robert seems to have married a Margaret Hobson in 1859:

If you look at the image, Robert is listed right next to a Joseph Scott age 40-50 who has 1 female in his household aged 20-30. Further down the same page James Scott age 20-30 also has in female in the household age 20-30. Possible Nancy was widowed and not actually born a Scott?

Sorry, just realized I had misread the marriage date. Joseph, James, and Elias Scott are possible family of Nancy.
The Pedigree at FamilySearch referenced in the Bio above is compiled from user submitted data. It is clear that the info for Robert's son Thomas is incorrect and conflated with another contemporary Thomas Stanley (very common name). As shown in Thomas's Bio here at WikiTree, it is clear that the son of Robert H. (Holmes appears to be speculative, and is likely a reference to the immigrant Ancestor Thomas's wife's name which is unknown, but listed in many online trees as Mary Holmes. There is no documented evidence of this name.) married Julia Ann Haynes and lived next door to his parents in 1850.
