John was the son of US Congressman John and (Unknown) Locke
He married first Jane Ermina Starkweather in 1829 - their children included John W., Charles Starkweather, Jane Ermina, Mary Haven, Grace LeBaron and Francis Goodwin Locke
After the death of wife Jane he married Harriet Brown Pinkham in 1859
He is buried at Lowell Cemetery, Lowell, Middlesex, Massachusetts, USA
