John was born in 1764, the son of Joseph and Hannah (Richmond) Andrews, in Higham, Suffolk County, Massachusetts, where he was most likely raised. John studied under and became an Ordained Colleague Pastor, with the Rev. Thomas Cary of the First Religion Society in Newburyport, Massachusetts on December 10, 1788.
On September 8, 1789, John married Margaret Wigglesworth, the daughter of Rev. Doctor Edward and Margaret (Hill) Wigglesworth, in Cambridge, Middlesex County, Massachusetts. John and Margaret settled in Newburyport where he held his ministry and had the following children:

Hannah Richmond Andrews (1795–1797)
John Andrews Jr. (1797–1874)
Hannah Richmond Andrews (1801–1876)
Mary Jane Andrews (1803–1893)
Of the children, only Edward and John Jr. married and only John Jr. had children.
John resigned his office as the Minister of the First Religion Society on May 1, 1830.
Margaret passed away in 1837 at 70 years of age while John departed this life in 1845 at 82 years old. Their bodies are buried together in the Oak Hill Cemetery in Newburyport, Essex County, Massachusetts.
