Carole Partridge has made an educated guess that it was Anne Hosmer, based on the English patterns of naming children around the 1600s. In most families, the first son was named after the father and the first daughter was named after the mother. In this family, the first son, this John, was named for his father, and the second child, a girl, was named Anne. Therefore, it is likely that the children's mother was also named Anne.
