William was born in 1792. He was the son of James Barlow and Elizabeth Ruxton.
He was educated by Mr Falloon and entered Trinity College Dublin on 1 June 1807. B.A. 1812, M.A. 1818. [1]
He was ordained Deacon 25 April 1815 (Kilmore) ; was Perpetual Curate Eglish, 1826-37; Vicar Carlingford, 1837-1852. He was collated Treasurer of Armagh and Rector of Creggan (Armagh) on 16 March 1852.
William married Catherine Disney, daughter of Thomas Disney and and Anne Purdon, on 5 May 1825.

Catherine had been in love with William Rowan Hamilton, but her family, believed him unsuitable, and she had been forced to marry the Rev William Barlow. The background and effects on Catherine are outlined by Anne van Weerden, in her study published in 2019, entitled Catherine Disney, A Biographical Sketch. The couple lived apart after Catherine attempted suicide in 1848. She died five years later in 1853.
William died in 1871. [2]
