Andrew Johnstone was born on March 13, 1805 in South Carolina, United States, son of William Andrew Johnston (1776–1840) and Anna Pinckney (~1778–~1853).
He married (1st) Sophia Beaumont Clarkson (1808-1845). They had one child (William Clarkson Johnstone). He married (2nd) Mary Barnwell Elliott (1824–1909).
Andrew Johnstone was the owner of Annandale Plantation (later renamed Millbrook) and was a rice planter in Georgetown County, South Carolina.

He sold Annandale in 1863 and retired to Beaumont in Flatrock, North Carolina where he had been a summer resident for over 20 years.
Louise Bailey reported the following in the Georgetown Times:
In June of 1864, deserters from Alabama came into the Johnstone home during dinner with the family. First they demanded food for their horses, then dinner and they were invited into the dining room. Elliott, Mr. Johnstone's 12 year old son, suspected something was amiss and got his gun. The men shot Andrew and Elliott shot back, wounding three of the men. Their intent was robbery as one of the men was found later with gold, silver, jewelry and Confederate money. Andrew died within the hour.[2]
