John was the son of Thomas Holbrook,[1] one of the first settlers of Sherborn, Massachusetts.[2]
John served as constable of Sherborn in 1697. He was a "sealer of leather" from 1705 to 1732. He was commissioner of assessments in Sherborn in 1715. He served as selectman of Sherborn for six years, and was representative in 1772.[1]
He married Silence Wood in about 1694. In circa that year, they built their house in Sherborn, Massachusetts, on land given to them by his father-in-law. The house was located on 69 Forest street; it was still standing in 1999.[3][4][2] Descendants of John and Silence continued to live in this house in 1981.[2] Their son, Jonathan, was known to have resided in the house.[1]

John died on February 28, 1740. His wife died on May 11, 1756.[1]
