Joseph Varnum was born on 7 May 1710 in Dracut, Middlesex, Massachusetts Bay Colony, son of Joseph Varnum and Ruth Jewett.
His siblings were:

- Samuel Varnum (1715–1797)
- John Varnum (1721–1786)
